葛根素配伍丹酚酸B防治大鼠急性心肌梗死的协同效应研究
Study on the Synergistic Effects of Puerarin Combined with Salvianolic Acid B in Preventing Acute Myocardial Infarction in Rats

Abstract
Objective:Guided by the compatibility theory of Traditional Chinese Medicine(TCM)and focusing on the optimal pairing within the"Pueraria-Salvia"herb couple,this study aimed to investigate the synergistic protective effect of the combination of puerarin(the main active component of Pueraria lobata)and salvianolic acid B(the primary water-soluble component of Salvia miltiorrhiza)a-gainst acute myocardial infarction(AMI)in rats.Methods:The rat AMI model was established via caudal vein injection of isoprotere-nol.The rats were randomly divided into the following groups,including control,model,captopril(positive control),puerarin mono-therapy(low,medium,and high doses),salvianolic acid B monotherapy,and puerarin with salvianolic acid B combination group.Therapeutic efficacy was comprehensively evaluated using electrocardiography(ECG),echocardiography,serum enzymatic markers lactate dehydrogenase(LDH),creatine kinase(CK),and histopathological examination hematoxylin-eosin(HE)and 2,3,5-triphe-nyltetrazolium chloride(TTC)staining of myocardial tissue.Results:Compared with the control group,the model group exhibited sig-nificantly impaired cardiac function,markedly elevated serum levels of LDH and CK,severe myocardial necrosis,and inflammatory cell infiltration observed in HE staining.All treatment groups showed varying degrees of recovery in cardiac function,decreased serum LDH and CK activities,and some improvement in cardiac pathology compared to the model group.While both puerarin and salvianolic acid B monotherapies were effective,the combination group demonstrated particularly remarkable effects.Specifically,the puerarin plus salvianolic acid B group showed significantly superior efficacy compared to either monotherapy group in rectifying ECG abnormali-ties,restoring cardiac function indices left ventricular ejection fraction(LVEF)and left ventricular fractional shortening(LVFS),re-ducing serum LDH and CK activities,and alleviating myocardial necrosis and inflammatory cell infiltration.Conclusion:Both puerarin and salvianolic acid B monotherapies exerted certain therapeutic effects on AMI in rats.Furthermore,their combination demonstrated a synergistic interaction,producing significantly enhanced protective effects against AMI.This study identified the"puerarin-salvianolic acid B"pair as one of a preferred combination within the"Pueraria-Salvia"herb couple for exerting cardiovascular protective effects.关键词
