Untargeted Metabolomic Analysis of Differential Metabolites in Immature and Mature Testicular Sertoli Cells of Pigs

Abstract
Ensuring the sperm quality of breeding boars is a crucial link in pig breeding.As somatic cells in the seminiferous tubules that directly contact male germ cells,testicular Sertoli cells not only pro-vide structural support and nutritional supply for spermatogenesis,but also secrete a variety of regulatory factors to precisely modulate the entire process of sperm production.Metabolomics can elucidate biological events and phenotypic characteristics of organisms.Liquid chromatography coupled with high-resolution mass spectrometry in untargeted metabolomics possesses the advantages of high sensitivity,strong selec-tivity and high throughput,enabling flexible selection of ionization modes and mass analyzers to compre-hensively acquire metabolite information.In this study,untargeted metabolomic sequencing was performed on testicular Sertoli cells of Shaziling pigs at 30 and 90 days of age.A total of 439 metabolites were detec-ted,among which 184 differentially expressed metabolites were screened out,including 108 downregulated and 76 upregulated metabolites.The expression of metabolites in Sertoli cells differed significantly with testicular maturation(P<0.05).A total of 59 signaling pathways were enriched via KEGG annotation,and the differential metabolites were mainly enriched in metabolic pathways and organismal system path-ways.By analyzing the differential metabolites,this study further explored the interaction between porcine testicular Sertoli cells and testis development,which provides a theoretical basis for revealing the molecu-lar mechanism by which Sertoli cells regulate testicular development and spermatogenesis.关键词
