痛风汤对大鼠湿热蕴结型急性痛风性关节炎模型影响
Effect of Tongfeng Decoction(痛风汤)on Rat Model of Acute Gouty Arthritis with Damp Heat Accumulation Type

Abstract
Objective To investigate the anti-inflammatory mechanism of Tongfeng Decoction(痛风汤)on acute gouty arthritis model with damp heat accumulation type in rats.Methods 120 SD rats(weighing 180-200 g,both male and female)were randomly divided into groups.The rat model of acute gouty arthritis with damp heat accumulation type was induced.After intervention with Tongfeng Decoction,the changes in the levels of IL-1β,IL-18 and TNF-α in the serum and joint fluid of each experimental group were measured.The mRNA expression of TLR4,NF-κB,NLRP3,and IκB in the articular cartilage,as well as the protein expression of NLRP3,IκB,NF-κB,and TLR4 in the articular cartilage were assessed.The articular cartilage was stained with HE to observe the cytological changes.Results The rat model of acute gouty arthritis with damp heat accumulation type was successfully induced.Compared with the control group,the levels of IL-1β,IL-18,and TNF-α in the serum of the model group rats were significantly increased(P<0.05);the mRNA expression of TLR4,NF-κB,and NLRP3 in the articular cartilage was significantly increased(P<0.05),and the expression of IκB mRNA was significantly decreased(P<0.01);the protein expression levels of TLR4,NF-κB,and NLRP3 in the articular cartilage were significantly increased,while the expression level of IκB protein in the articular cartilage of the model group rats was significantly decreased.Compared with the model group and the positive control group,the above indicators in each experimental group showed varying degrees of improvement.Meanwhile,the HE staining results of the rat articular cartilage showed that compared with the model group,the tissue structure and morphology of the articular cartilage in the Tongfeng Decoction group and the positive control group tended to normalize,and the pathology was significantly improved.Conclusion Tongfeng Decoction has a significant anti-inflammatory effect on acute gouty arthritis with damp heat accumulation type in rats.关键词
