Microfluidics-based Integrated Analysis of Single-Cell Transcriptomics and Proteomics

Abstract
Single-cell multi-omics has become a crucial frontier in life sciences,providing powerful tools for investigating cellular heterogeneity and complex biological processes.Compared with single-omics analysis,the integration of single-cell transcriptomics and proteomics enables a more comprehensive characterization of the dynamic relationship between gene expression and cellular functional states,thereby offering multidimensional molecular information for understanding disease mechanisms.Benefiting from nanoliter-scale reaction systems and precise fluid manipulation,microfluidic technology has played an essential role in the development of integrated single-cell multi-omics analysis.In recent years,a series of microfluidics-based strategies have been developed for simultaneous analysis of single-cell transcriptomes and proteomes.According to the roles of microfluidic technologies in these approaches,the integration strategies can be categorized into three types:component separation strategies,barcode-assisted synchronous library construction strategies,and spatial labeling strategies.This review summarizes recent advances in microfluidics-based integrated single-cell transcriptomic and proteomic analysis from the perspectives of analytical principles,technical characteristics,as well as their advantages and limitations.Finally,the key technical challenges and future development trends in this field are discussed.关键词
