基于16S rRNA宏基因组技术探究口腔白斑病的微生物群落特点
Characterization of the microbial community in oral leukoplakia using 16S rRNA metagenomic sequencing
摘要
Abstract
Objective:To analyze the characteristics of the microbial community in oral leukoplakia(OLK)and the differences in microbial distribution between OLK with and without epithelial dysplasia, thereby exploring the potential role of the oral microbiota in the malignant transformation of OLK.Methods:Patients with histologically confirmed OLK,both with and without epithelial dysplasia, were enrolled.Mucosal swabs were collected from the lesions.Microbial sequencing and anal-ysis of the collected oral samples were performed using 16S rRNA metagenomic technology.Oral samples from healthy indi-viduals served as controls.Differences in microbial diversity and composition at the phylum and genus levels were compared among the groups. Results:Compared with the healthy control group, the microbial diversity in OLK lesions was significantly reduced(Chao index,P<0.001),and the microbial composition was markedly different. In terms of microbiota composition, genera such as Neisseria,Prevotella_7,and Campylobacter were significantly enriched in OLK lesions, while Haemophilus and Streptococcus were significantly reduced.Of particular note, the microbial composition differed between OLK with dysplasia and OLK without dysplasia. The former showed a significant increase in Fusobacterium,Stenotrophomonas,Aggregatibacter, and Porphyromonas compared to the latter. Conclusion:This study reveals the characteristics of microbial community altera-tions in oral leukoplakia lesions. It provides new clues for in-depth research into the pathogenesis and malignant transforma-tion mechanisms of OLK,and also offers new insights for the prevention and treatment of OLK carcinogenesis.关键词
