Lipid metabolic reprogramming in immune regulation of sepsis-associated organ injury:Current frontier
摘要
Abstract
Sepsis is a syndrome characterized by a dysregulated host response to infection, with progressive organ dysfunction as its core feature. The disease course exhibits a dynamic interplay or sequential transition between an early hyperinflammatory response and subsequent immunosuppression.Recent advances in immunometabolism have revealed that lipid metabolism not only participates in energy supply and cell membrane structural integrity but also profoundly regulates immune responses through multiple signaling pathways, including the synthesis of lipid mediators, membrane lipid raft-mediated signal transduction, lipid droplet formation, and lipid peroxidation.Furthermore, abnormal lipid metabolism is closely associated with endothelial barrier disruption, microcirculatory dysfunction, and regulated cell death, collectively contributing to the development and progression of multiple organ dysfunction. This article systematically summarizes the mechanisms underlying the roles of key pathways—including fatty acid metabolism, cholesterol homeostasis, the sphingolipid-sphingosine-1-phosphate signaling axis, lipid droplet dynamics, and pro-resolving lipid mediators—in the immune regulation and organ injury associated with sepsis. It also emphasizes the differences based on cell type, disease stage, and organ specificity, and further explores potential biomarkers and targeted intervention strategies.关键词
