HPLC法同时测定大豆杜仲淫羊藿颗粒中大豆异黄酮和淫羊藿苷的含量
Simultaneous Determination of Soybean Isoflavones and Icariin in Soybean-Eucommia-Epimedium Granules by HPLC

Abstract
To establish a method for the simultaneous determination of soy isoflavones(daidzin,genistin,daidzein and genistein)and icariin in Soybean-Eucommia-Epimedium Granules.In this study,high-performance liquid chromatography was employed.A Shimadzu Shim-pack GWS C1 8 reversed-phase column(5 μm,4.6 mm × 250 mm)was used.Gradient elution was performed with acetonitrile(A)and phos-phoric acid aqueous solution(pH=3,B)as the mobile phase.The injection volume was 10 μL,the column temperature was maintained at 30℃,and the detection wavelength was set at 260 nm.The results indicated that daidzin,genistin,daidzein,genistein,and icariin showed good linear relationships between their contents and peak areas within the ranges of 7.94～63.52 μg/mL,9.28～74.24 μg/mL,4.84～38.72 μg/mL,2.22～17.72 μg/mL,and 2.516～314.5 μg/mL,respectively.The correlation coefficients(R2)were 0.999 9,0.999 9,0.999 2,0.999 1,and 0.999 9,respectively.In the precision test,the relative standard deviations(RSDs)of the five components were 0.362％,0.358 8％,1.251 5％,0.220 3％,and 0.357 7％,respectively.The stability test demonstrated that all components in the sample solution remained stable within 24 h,with RSDs of 0.665 2％,1.835 6％,1.423 9％,0.762 6％,and 0.555 8％,respectively.The average recoveries were 101.70％,100.66％,102.67％,99.40％and 102.29％,respectively.The average contents of daidzin,genistin,daidzein,genistein,and icariin in three batches of products determined by this method were(39.79±0.75)mg/100 g,(32.52±0.37)mg/100 g,(20.04±0.337 8)mg/100 g,(9.48±0.15)mg/100 g,and(371.18±2.16)mg/100 g(n=9),respectively.The established method is accurate,simple,and reliable,and can be applied for the con-tent determination of Soybean-Eucommia-Epimedium Granules.关键词
