Abstract
Vepdegestrant is an oral heterobifunctional protein degrader.It is a small molecule comprised of an estrogen receptor binding domain joined by a linker to an E3 ubiquitin ligase binding domain.Vepdegestrant is indicated for the treatment of adults with estrogen receptor(ER)-positive,human epidermal growth factor receptor 2(HER2)-negative,estrogen receptor 1(ESR1)-mutated advanced or metastatic breast cancer,as detected by a U.S.Food and Drug Administration(FDA)-authorized test,with disease progression following at least one line of endocrine therapy.Clinical trials have shown that vepdegestrant prolonged progression-free survival(PFS)versus fulvestrant in patients with ESR1-mutated ER-positive/HER2-negative advanced breast cancer(5.0 vs 2.1 months).This paper presents an overview of the mechanism of action,pharmacokinetic characteristics,clinical efficacy,safety profile and clinical positioning of vepdegestrant.关键词
