传染性法氏囊病病毒感染组织和细胞中chApoA-Ⅰ表达检测
Detection of chApoA-Ⅰ expression in infectious bursal disease virus-infected tissues and cells
摘要
Abstract
Apolipoprotein A-Ⅰ(ApoA-Ⅰ),the principal protein component of high-density lipopro-tein(HDL),plays a critical role in lipid transport, inflammation, tumor progression, and viral infec-tion.Previous differential proteomics revealed that chicken ApoA-Ⅰ(chApoA-Ⅰ)was upregulated in chicken embryo fibroblasts infected with infectious bursal disease virus(IBDV).To further character-ize the expression patterns of chApoA-Ⅰ in tissues and cells during IBDV infection,real-time quanti-tative PCR(RT-qPCR)and Western blot analysis were applied to assess the mRNA and protein expression levels of ApoA-Ⅰ in the heart, liver, spleen, bursa of Fabricius, thymus, and lung tissues of IBDV-infected chickens,as well as in IBDV-infected DF-1 and HEK-293T cells. The results showed that IBDV infection in-duced significant upregulation the relative mRNA and protein expression levels of chApoA-Ⅰ in the spleen, bursa, and thymus,as well as in the heart. However, only protein expression levels were elevated in the lung and liver tissues without corresponding changes at the mRNA expression level. Moreover, both mRNA and protein expression levels of ApoA-Ⅰ were significantly upregulated in IBDV-infected DF-1 and HEK-293T cells.Further analysis comparing the molecular weights of chApoA-Ⅰ expressed in infected DF-1 cells with the full-length,signal peptide-cleaved precursor, and mature forms suggested that chApoA-Ⅰ predominantly existed in either the precursor or mature form during IBDV infection. These findings provide the experimental evidence for further investigation into the functional roles and mechanism of chApoA-Ⅰ in IBDV replication and pathogenesis.关键词
