Quality evaluation of Uncariae Ramulus cum Uncis based on UPLC fingerprints,chemometrics,content determination and chromatic values

Abstract
AIM To evaluate the quality of Uncariae Ramulus cum Uncis.METHODS The UPLC fingerprints were established,after which cluster analysis,principal component analysis and orthogonal partial least squares discriminant analysis were performed,HPLC was adopted in the content determination of isocorynoxeine,corynoxeine,isorhynchophylline and rhynchophylline,colorimetric method was applied to determining the content of total flavonoids,the comprehensive evaluation was performed by CRITIC-TOPSIS method,ΔL*,Δa*,Δb* andΔE*ab were determined,and the relationships between active constituent contents and chromatic values.RESULTS There were 10 common peaks in the fingerprints for 38 batches of medicinal materials with the similarities of 0.330-0.977,8 of which were identified.Various batches of medicinal materials were clustered into 3 types,4 principal components demonstrated the accumulative variance contribution rate of 77.937％,chlorogenic acid and isochlorogenic acid B were taken as potential quality markers.The average total content of 4 alkaloids in medicinal materials produced in Guizhou was the highest,while that produced in Guangxi was the lowest;the content of total flavonoids in the S02 medicinal material was the highest,while that in the S21 medicinal material was the lowest.The medicinal materials produced in Guangxi,Hunan,Guizhou and Chongqing displayed high rankings.The color differences were mainly reflected in brightness and yellow-blue tone,and flavonoids and phenolic acids might be the main material basis of color formation.CONCLUSION This experiment characterizes and quantifies Uncariae Ramulus cum Uncis color,and combines it and active constituent contents,which can comprehensively reflect the quality differences of this medicinal material from different producing areas,and provide a reference for the improvement of its quality evaluation.关键词
