PD-L1与STAT1在弥漫性大B细胞淋巴瘤中的表达及预后研究
The expression and prognosis of PD-L1 and STAT1 in diffuse large B-cell lymphoma

Abstract
This study aims to investigate the expression and correlation of PD-L1 and STAT1 in DLBCL,and to evaluate their association with the clinicopathological features and prognosis of DLBCL patients.The transcriptional expression profiles of PD-L1 and STAT1 in DLBCL tissues were analyzed using the GEPIA and GEO public databases.Additionally,immunohistochemistry was performed to detect the protein expression of PD-L1 and STAT1 in tumor tissues from 72 DLBCL patients,whose clinical data were also collected.Combined with the survival analysis and multivariate Cox regression analysis,these data were used to validate the prognostic value of PD-L1 and STAT1 in DLBCL.Consistent results across GEPIA,GEO,and immunohistochemical cohorts uniformly showed that both PD-L1 and STAT1 were significantly upregulated in DLBCL tissues(P<0.05).Among the 72 DLBCL patients,the positive expression rates of PD-L1,STAT1,and PD-L1/STAT1 co-expression were 69.44%,77.78%,and 56.94%,respectively.PD-L1 expression was positively correlated with STAT1 expression(r=0.256 7,P=0.029 5).Moreover,PD-L1/STAT1 co-expression was significantly associated with poor prognosis in DLBCL patients(P<0.001).Further multivariate Cox regression analysis showed that positive expression of PD-L1 and STAT1 remained independent adverse risk factors for both OS and PFS(PD-L1:P<0.001 for OS and PFS;STAT1:OS,P=0.032;PFS,P=0.014).This study confirms that PD-L1 and STAT1 are co-upregulated in DLBCL,and that their individual positive expression as well as co-expression can serve as independent molecular markers for evaluating the prognosis of DLBCL patients.These findings suggest that PD-L1 and STAT1 may represent potential therapeutic targets for DLBCL,providing important theoretical and experimental support for individualized prognostic stratification and the development of targeted treatment strategies in clinical practice.关键词
