西藏波氏琵蝎防御素SPDef4的制备与抗菌功能研究
Preparation and antimicrobial function research of a defensin polypeptides SPDef4 from Scorpiops pococki in Xizang

Abstract
This study was designed to produce the defensin peptide SPDef4(Scorpiops pococki defensin 4)via prokaryotic expression,evaluate its antimicrobial activity,and preliminarily explore its mechanism of action.SPDef4 was recombinantly expressed using an Escherichia coli expression system.The minimum inhibitory concentration(MIC)of SPDef4 was determined using the broth dilution method.Preliminary elucidation of antibacterial mechanisms via DMAO/PI dual staining in conjunction with confocal and transmission electron microscopy(TEM).Recombinant SPDef4 exhibits antibacterial activity against the Gram-positive bacterium Staphylococcus aureus,with no efficacy observed against the Gram-negative bacterium Escherichia coli.DMAO/PI double staining and transmission electron microscopy results suggest that SPDef4 exerts its antibacterial effect by disrupting cell membrane integrity.The defensin SPDef4 emerges as a novel member of scorpion-derived antimicrobial peptides with notable antibacterial activity.Future investigations should expand its antibacterial spectrum assessment and explore its potential as a broad-spectrum antimicrobial agent.关键词
