皂翘消癖方提取工艺优化及其颗粒制备工艺研究
Optimization of extraction process and granule preparation process of Zaoqiao Xiaopi Formula(皂翘消癖方)
摘要
Abstract
Objective To optimize the extraction process of Zaoqiao Xiaopi Formula(皂翘消癖方)and estab-lish the formulation preparation method and quality control method for Zaoqiao Xiaopi Granules(皂翘消癖颗粒).Methods An orthogonal experimental design was employed,using the comprehensive score of phillyrin extraction rate and dry extract yield as evaluation indicators to optimize the water extraction parameters of Zaoqiao Xiaopi For-mula.A multi-index comprehensive evaluation method was adopted,with granule formability,appearance,particle size,solubility,moisture absorption rate,and phillyrin content as examination indicators,to screen the type,ratio,and amount of excipients and determine the optimal preparation process.High-performance liquid chromatography(HPLC)was used to determine the content of indicator components and establish a quality control method.Results The optimal extraction process for Zaoqiao Xiaopi Formula was as follows:Curcuma zedoary was first subjected to steam distillation to extract volatile oil,and the residue was mixed with the other prescribed medicinal materials,adding 6 times the amount of water,decocted once for 1 hour.The optimal preparation process for Zaoqiao Xiaopi Granules was:using a composite excipient of dextrin-soluble starch(2∶3),with a dry extract powder to composite excipient mass ratio of 1.0∶1.5,adding the volatile oil of Curcumae Rhizoma and mixing evenly,followed by wet granulation and drying at 40 ℃ for 24 h.The preparation results of three batches of samples showed that the gra-nules had good appearance and stable phillyrin content,meeting the relevant requirements of the Chinese Pharmaco-poeia(2020 Edition,Volume Ⅳ)for granules.Conclusion The extraction and preparation processes established in this study are reasonable with good reproducibility,and the quality control method is simple and reliable,providing experimental evidence for the subsequent development and industrial production of Zaoqiao Xiaopi Granules.关键词
