Expression levels of SOX7 and CSMD1 in endometrial cancer tissues and their associations with pathological characteristics and prognosis

Abstract
Objective To investigate the expression levels of SRY-box related gene 7(SOX7)and CUB and Sushi multiple domains 1(CSMD1)in endometrial cancer(EC)tissues and their associations with pathological features and prognosis.Methods A total of 125 EC patients admitted to the First Affiliated Hospital of Hebei North University from November 2019 to January 2022 were selected.Intraoperative EC tissue specimens(EC group)and corresponding adjacent non-cancerous tissues specimens(adjacent group)were collected.According to patient prognosis,they were divided into the poor prognosis group(n=52)and the good prognosis group(n=73);qRT-PCR was used to detect SOX7 and CSMD1 mRNA levels;immunohistochemistry was used to detect SOX7 and CSMD1 expression;Kaplan-Meier survival analysis was performed to evaluate patient prognosis;multivariate Cox regression analysis was used to analyze influencing factors,and receiver operating characteristic(ROC)curves were used to evaluate predictive value.Results The mRNA expression levels of SOX7 and CSMD1 in the EC group were lower than those in the adjacent non-cancerous group,and the differences were statistically significant(t=24.538 and 24.034,respectively,P<0.05);the positive protein expression rates of SOX7 and CSMD1 in the EC group were both lower than those in the adjacent non-cancerous group,with statistically significant differences(χ2=13.581 and 13.511,respectively,P<0.05);the negative expression of SOX7 and CSMD1 in the EC group was associated with high FIGO stage,poor differentiation,and lymph node metastasis(χ2-values ranged from 6.108 to 10.523,P<0.05);the 3-year survival rate of patients positive for SOX7 and CSMD1 was higher than that of patients with negative expression(χ2=4.195 and 12.755,respectively,P<0.05);multivariate Cox regression showed that high FIGO stage,low differentiation,and lymph node metastasis were risk factors affecting patient prognosis(HR=2.304,3.048,and 3.521,respectively,P<0.05),while high mRNA expression of SOX7 and CSMD1 were a protective factors(HR=0.324 and 0.304,respectively,P<0.05);ROC curve analysis showed that the AUC for predicting EC patient prognosis using combined SOX7 and CSMD1 mRNA expression was 0.840,which was superior to prediction using either alone(Z=2.715 and 2.718,respectively,P<0.05).Conclusion The expression levels of SOX7 and CSMD1 were abnormally downregulated in EC tissues,which were related to the pathological characteristics and prognosis of patients,and the combined detection could improve the predictive value for patient prognosis.关键词
