Effects of Different Storage Temperatures and Drying Treatments on Pollen Viability of Dactylorhiza hatagirea

Abstract
Dactylorhiza hatagirea as one species of the authentic edible and medicinal orchid"shouzhangshen"has good economic value and development potential.To explore the effect of storage temperature and drying treatment on the pollen vitality of D.hatagirea,this study conducted three storage temperatures(-20℃,4℃,25℃)and anhydrous silica gel drying treatment on it.The pollen vitality after 30 days,60 days,and 90 days of storage was determined using the TTC staining method.This result indicates that within the experimental range,the storage temperature is negatively correlated with the pollen vitality of D.hatagirea,and drying treatment is beneficial for maintaining the pollen vitality of D.hatagirea;and low temperature(-20℃)has a significant effect on delaying the decrease in pollen vitality of D.hatagirea,but drying treatment is only suitable for short-term storage for 30 days.Therefore,to ensure the pollen quality of D.hatagirea,it is recommended to dry it and store it in a low-temperature refrigerator at-20℃for no more than 30 days.关键词
