Abstract
Objective:To explore the effect of Poria cocos polysaccharide on gastric mucosal injury in rats with chronic atrophic gastritis(CAG)and explore its potential mechanism involving the interleukin(IL)-10(IL-10)/JAK1/STAT3 signaling pathway. Methods:SD rats were randomly divided into the following groups:control, model,Poria cocos polysaccharide,Poria cocos polysaccharide+RO8191(a JAK/STAT3 signaling pathway activator),and Poria cocos polysaccharide+IL-10 neutralizing antibody.A CAG rat model was established by using N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)combined with irregular feeding. After intervention with Poria cocos polysaccharides, the pH value of rat gastric juice was measured.ELISA was applied to measure serum levels of gastric function-related indicators and inflammatory cytokines.H-E staining was performed to evaluate gastric mucosal histopathological change.Immunohistochemical staining was employed to detect the numbers of G cell and D cell in the gastric mucosa of rats.TUNEL staining was applied to detect apoptosis of gastric mucosal tissue cells of rats.Western blotting was adopted to detect the IL-10/JAK1/STAT3 signaling pathway-related proteins in the gastric mucosal tissues of rats. Results:Compared with the control group, the model group showed significant pathological damage in gastric mucosa, with increased gastric juice pH value, gastric mucosal tissue cell apoptosis index,IL-8 and IL-6 levels, and elevated p-JAK1/JAK1 and p-STAT3/STAT3 ratios, while gastrin, motilin,G cell and D cell optical density values, and serum and tissue protein expression of IL-10 were decreased.Compared with the model group, the Poria cocos polysaccharide group exhibited alleviated gastric mucosal damage, decreased gastric juice pH value, gastric mucosal tissue cell apoptosis index,IL-8 and IL-6 levels, and p-JAK1/JAK1 and p-STAT3/STAT3 ratios, along with increased gastrin, motilin,G cell and D cell optical density values, and IL-10 expression.Compared with the Poria cocos polysaccharide group, both the Poria cocos polysaccharide+RO8191 and the Poria cocos polysaccharide+IL-10 neutralizing antibody groups showed aggravated pathological damage to gastric mucosal tissue, increased gastric juice pH value, gastric mucosal tissue cell apoptosis index,IL-8 and IL-6 levels, and p-JAK1/JAK1 and p-STAT3/STAT3 ratios, while decreased gastrin, motilin,G cell and D cell optical density values, and IL-10 serum and protein expression. Conclusion:Poria cocos polysaccharide can alleviate gastric mucosal injury in CAG rats, possibly by modulating the IL-10/JAK1/STAT3 signaling pathway.关键词
