1株羊肺源肺炎克雷伯菌的分离鉴定及其耐药性、毒力分析
Isolation,identification,drug resistance,and virulence analysis of one strain of Klebsiella pneumoniae isolated from sheep lung
摘要
Abstract
The study aimed to understand the causes of coughing and death in sheep at a large-scale sheep farm in Yutian County,Xinjiang,and to provide references for the prevention and control of related diseases.The experiment conducted bacterial isolation and purification of lung tissue samples from the deceased sheep,and carried out 16S rRNA,biochemical identification,and specific gene and serotype PCR tests.The K-B method was used to detect the sensitivity of the isolates to antibacterial drugs,and PCR was used to detect the drug resistance and virulence genes carried by them.Subsequently,a mouse challenge test was conducted to evaluate the pathogenicity.The results showed that one strain of Klebsiella pneumoniae was isolated and identified from five lung tissue samples of the deceased sheep,named FKP1,belonging to the K1 serotype,and was in the same phylogenetic branch as the Klebsiella pneumoniae N1-T-2 strain.The isolates were resistant to amoxicillin,vancomycin,penicillin,gentamicin,and ampicillin,carried multiple drug resistance(blaSHV,blaTEM,oqxA,gyrA)and virulence(fimH,mrkD,uge,ompA,ureA,irp2,wabG)genes.When the bacterial solution concentration of the isolates was 2.6×109 and 2.6×108 CFU/mL,100%of the mice were killed,and the median lethal dose(LD50 value)was 2.8×106 CFU.The study showed that through the experimental isolation,one strain of K1 serotype Klebsiella pneumoniae was obtained,which was resistant to β-lactam drugs and carried multiple drug resistance and virulence genes.It exhibited strong pathogenicity in mice.关键词
