Isolation, identification, and probiotic characterization of equine lactic acid bacteria
摘要
Abstract
To explore the intestinal lactic acid bacteria resources of native horse breeds in China, fresh fecal samples from Shandan horses in Zhangye were used in this study.Selective culture combined with 16S rDNA molecular technology was applied for bacterial isolation and identification. The in vitro probiotic properties of the isolates, including acid tolerance, bile salt tolerance,auto-aggregation,surface hydrophobicity and antibacterial activity, were systematically evaluated. The results showed that a total of three lactic acid bacterial strains were isolated.Strain Z13 was identified as Limosilactobacillus equigenerosi,Z14 as Limosilactobacillus equi, and Z15 as Pediococcus pentosaceus. The three isolates exhibited obvious strain specificity in probiotic performance. After exposure at pH 3.0 for 4 h,the survival rate of strain Z13 reached 113.10%,showing the best acid tolerance;it also had a high hydrophobicity rate(77.06%)and auto-aggregation rate(30.19%),indicating excellent potential for intestinal colonization.Strain Z14 presented the highest tolerance to 0.5%bile salt and displayed the strongest antibacterial activity against Staphylococcus aureus,Escherichia coli and Salmonella.Strain Z15 showed relatively weaker overall performance. This study demonstrates that isolates Z13 and Z14 derived from the intestinal tract of Shandan horses possess respective superiorities in tolerance, adhesion capacity and antibacterial activity, which can serve as promising candidate strains for the development of indigenous livestock and poultry microecological preparations with strong adaptability and stable functions.关键词
