甲磺酸去铁胺缓解小鼠实验性结肠炎的效果及机制研究
Ameliorating Effect and Mechanism of Deferoxamine on DSS-induced Ulcerative Colitis in Mice

Abstract
The aim of this study was to investigate the ameliorative effect and mechanism of Deferoxamine Mesylate(DFO)on DSS in-duced ulcerative colitis in mice.Forty female C57BL/6J mice were randomly assigned to four groups(n=10),i.e.,blank control group(NS),DFO drug group(DFO),ulcerative colitis model group(DSS),and DFO treated group(DSS+DFO).After a week of acclimatization of mice,mice in the DFO group and DSS+DFO group were given an intraperitoneal injection of 10 mg/kg DFO in the second week,and mice in the DFO group and DSS+DFO group were given 3.5%DSS for modeling in the third week.The body weight,fecal consistency,and bleeding were recorded for each mouse during modeling.After the establishment of the model,the mice were sacrificed after venous blood collection,and the blood,liver as well as colon tissues were collected for subsequent test de-tection.After DFO treatment,compared to the DSS group,the clinical symptoms were effectively relieved,and the DAI decreased.Pathological sections showed that DFO improved the pathological changes of colon tissue,the intestinal wall edema was improved,the intestinal gland and goblet cells were recovered,and large areas of mucosal epithelial cells were improved.After DFO treatment,the expression of IL-1β and TNF-α in serum of UC mice was inhibited,and the transcription of I L-1 β,I L-6,N L R P3 and NF-κB in colon tissue was down-regulated.DSS could relieve the clinical symptoms of DSS induced ulcerative colitis and effectively inhibit the inflammatory response.Its mechanism of action involved the inhibition of IL-1β and TNF-α and was related to the transcription regu-lation of NF-κB and NLRP3 in colon.关键词
