PRDX2对子痫前期滋养层细胞增殖和侵袭的影响
Effects of PRDX2 on the proliferation and invasion of trophoblasts in preeclampsia

Abstract
Objective To investigate the effects and mechanisms of peroxiredoxin 2(PRDX2)on the proliferation and inva-sion of trophoblast cells in preeclampsia.Methods The human trophoblast cell line HTR-8/Svneo was used to construct a PRDX2 overexpression model.Cells were randomly divided into the oe-NC group(transfected with empty plasmid),oe-PRDX2 group(transfected with PRDX2-overexpression plasmid),and oe-PRDX2+PNU-74654 group(transfected with PRDX2-overex-pression plasmid and treated with the Wnt/β-catenin pathway-specific inhibitor PNU-74654).qRT-PCR and Western blot were used to detect the mRNA and protein expression levels of PRDX2.Western blot was used to detect the expression of Wnt3a and β-cate-nin as key proteins of the Wnt/β-catenin signaling pathway.The effects of PRDX2 overexpression and its combination with PNU-74654 on cell proliferation and invasion were evaluated by CCK-8 and Transwell assays,respectively.Results Compared with the oe-NC group,the oe-PRDX2 group showed significantly increased mRNA and protein expression levels of PRDX2,cell proli-feration activity,number of invasive cells,and expression of Wnt3a and β-catenin(t=7.36-13.11,P＜0.01).There were signifi-cant differences in cell proliferation and invasion among the three groups(F=31.30,37.61,P＜0.01).Specifically,proliferation and invasion were significantly higher in the oe-PRDX2 group than in the oe-NC group,while they were significantly lower in the oe-PRDX2+PNU-74654 group than in the oe-PRDX2 group(P＜0.01).Conclusion PRDX2 overexpression may promote the proliferation and invasion of trophoblast cells by activating the Wnt/β-catenin signaling pathway.关键词
