Effect of the replication factor C subunit 4 gene on the proliferation,invasion,and migration of triple-negative breast cancer cells

Abstract
Objective To investigate the expression of the replication factor C subunit 4(RFC4)gene in triple-negative breast cancer(TNBC)cells and its effect on the proliferation,migration,and invasion of TNBC cells.Methods The bioinfor-matics method was used to validate the expression level of RFC4.Western blot was used to measure the expression level of RFC4 in TNBC cell lines MDA-MB-231,MDA-MB-468,and BT-549,as well as the knockdown and overexpression efficiency of the RFC4 gene;colony formation assay was used to measure cell proliferation,and Transwell assay was used to measure cell migration and invasion.Results The bioinformatics analysis showed that RFC4 was highly expressed in the high-throughput sequencing and microarray data of breast cancer.RFC4 had a high expression level in MDA-MB-231 and MDA-MB-468 cells and a low expres-sion in BT-549 cells.After the knockdown of the RFC4 gene in MDA-MB-231 and MDA-MB-468 cells,there were significant re-ductions in the number of cell migration and invasion(t=23.50-56.10,P＜0.01)and the proliferation ability of cells(t=24.46,17.85,P＜0.01).After the overexpression of the RFC4 gene in BT-549 cells,there were significant increases in the number of cell migration and invasion(t=7.40,18.20,P＜0.01)and the proliferation ability of cells(t=15.29,P＜0.01).Conclusion Inhibi-tion of RFC4 gene expression can inhibit the proliferation,migration,and invasion of TNBC cells,while increasing RFC4 gene ex-pression has the opposite effect.关键词
