Association of the risk of atherosclerotic cardiovascular disease with biomarkers for Alzheimer's disease and cognitive function

Abstract
Objective To investigate the association of the risk of atherosclerotic cardiovascular disease(ASCVD)with cere-brospinal fluid(CSF)biomarkers for Alzheimer's disease(AD)and cognitive function.Methods A total of 1 371 non-dementia subjects were selected from the Chinese Alzheimer's Biomarker and Lifestyle(CABLE)study cohort.The 10-year ASCVD risk score was calculated using the American College of Cardiology/American Heart Association(ACC/AH A)Risk Estimator to pre-dict the risk of cardiovascular disease in the future,and according to the 10-year ASCVD risk score,the subjects were divided into very low risk group with 287 subjects,low risk group with 255 subjects,moderate risk group with 371 subjects,and high risk group with 458 subjects.The multiple linear regression model was used to analyze the association of 10-year ASCVD risk score with AD biomarkers in CSF(amyloid-β42(Aβ42),total tau(T-tau),and phosphorylated tau(P-tau)),Mini-Mental State Examination(MMSE)score,and Montreal Cognitive Assessment(MoCA)score,and the mediation effect analysis was used to assess whether CSF AD biomarkers mediated the association between ASCVD risk and cognitive function.Results The 10-year ASCVD risk score was positively associated with CSF P-tau(β=0.199,P＜0.001),CSF T-tau(β=0.269,P＜0.001),CSF T-tau/Aβ42(β=0.132,P＜0.001),and CSF P-tau/Aβ42(β=0.069,P＜0.05),was not significantly associated with CSF Aβ42(β=0.037,P＞0.05),and was negatively associated with MMSE score(β=-0.168,P＜0.001)and MoCA score(β=-0.240,P＜0.001).The mediation effect analysis showed that tau-related biomarkers partially mediated the effect of ASCVD risk on cognitive function.Conclusion The 10-year ASCVD risk score is positively associated with CSF T-tau and P-tau in the non-dementia population,sug-gesting that the risk of cardiovascular diseases may impair cognitive function through the tau-related pathways.关键词
