Current status of long-acting GLP-1 receptor agonists development

Abstract
Obesity and diabetes have emerged as major global challenges in the prevention and control of metabolic diseases.Native glucagon-like peptide-1(GLP-1)possesses an extremely short circulating half-life,necessitating long-acting modifications to enable its clinical application.This review systematically summarized recent advances in long-acting GLP-1 receptor agonists(GLP-1 RAs).At the molecular structural modification level,we focus on fatty acid chain conjugation for albumin affinity,molecular size expansion,and optimization strategies based on receptor pharmacology.At the formulation strategy level,we outline the current applications and development trends of delivery systems such as PLGA microspheres,in situ gels,and lipid carriers.Finally,we summarize the advantages and bottlenecks of existing long-acting technologies and provide perspectives on future research directions and clinical translation pathways for long-acting GLP-1 RA formulations.关键词
