Induction and proliferation of Gardenia jasminoides callus and its metabolites
摘要
Abstract
Pericarp, seeds and seed masses from Gardenia jasminoides fruits were evaluated as explants for callus induction. After optimizing the callus proliferation conditions,we compared the corresponding effects of solid culture flasks, liquid shake flasks and plant bioreactors on the proliferation of G.jasminoides callus.Liquid chromatography-mass spectrometry(LC-MS)was used to detect and analyze metabolites in G.jasminoides callus. As a result, the seed mass isolated from G.jasminoides fruit exhibited the highest callus induction efficiency, reaching 20.95%.The optimal medium for callus proliferation was MS medium supplemented with 1.5 mg/L 6-benzylaminopurine(6-BA),1.5 mg/L naphthaleneacetic acid(NAA),and 30 g/L sucrose.G.jasminoides callus displayed robust proliferation in solid culture flasks, shake flasks and plant bioreactors.A total of 1 281 metabolites were annotated in G.jasminoides callus through the screening with the COCOUNT database, including terpenoids, alkaloids, organic acids, amino acids and peptides, shikimates, phenylpropanoids and other classes.Overall,an efficient proliferation culture system was constructed for G.jasminoides callus, and the subsequent metabolomic profiling revealed that the callus accumulates a diverse array of bioactive natural constituents. Our study provides essential technical support for the resource-oriented utilization of G.jasminoides cell cultures.关键词
