Abstract
Objective To investigate the feasibility of electric nanomicro-needle introduction of testosterone propionate solution to construct androgenic alopecia model mice. Methods C57/BL6J male mice were randomly divided into four groups, namely the blank group, the microneedle group, the drug-coated group, and the subcutaneous injection group, and testosterone propionate solution was given to each group by the corresponding percutaneous administration,to construct a mouse model of androgenic alopecia.By observing the changes in skin color, hair growth timing, and hair coverage rate of the mice in each group,as well as conducting HE staining on the mouse skin tissues and examining the tissue morphology of the model mice under an electron microscope to calculate the hair follicle density per unit area;testing the levels of testosterone(T)and dihydrotestosterone(DHT)in the skin and blood of the mice in each group to explore the modeling effects. Results On the 7th day the hairs in the blank group group gradually grew out of the skin surface, the time for the hairs to grow out of the epidermis became longer in the subcutaneous injection group, the drug-coated group, and the microneedle group compared with the blank group, and all of them were statistically significant(P<0.05),on the 15th day the hairs in the blank group group had completely grown out,and the rate of growth of the hairs was slower in all of them in the subcutaneous injection group, the drug-coated group and the microneedle group compared with the blank group. On the 21st day the hairs in the subcutaneous injection group, the drug-coated group, and the microneedle group did not completely grow out,and there was a statistically significant difference in the rate of growth of the hairs in the subcutaneous injection group, the drug-coated group, and the microneedle group compared with the blank group(P<0.05).statistical significance(P<0.01).T and DHT in the skin and serum of all modeling groups were significantly higher compared with the the blank group group, and the levels of T and DHT in the skin and serum of the the subcutaneous injection group and the microneedle group groups were extremely statistically significant compared with the blank group(P<0.001).Conclusion The use of motorized microneedle to introduce testosterone propionate solution into the skin of mice may promote the miniaturization of hair follicles by increasing the levels of T and DHT in the mice's serum and skin, thereby constructing a mouse model of androgenetic alopecia.关键词
