Effect of Fire Needle Therapy Combined with Isotretinoin Soft Capsules on Inflammation and NLRP3/Caspase-1 Signaling Pathway in Acne Model Mice

Abstract
Objective To explore the effect of fire needle therapy combined with isotretinoin acid soft capsule on inflammation and NLRP3/Caspase-1 signaling pathway in acne model.Methods 60 mice were randomly divided into 6 groups,10 in each group:control group,model group,fire needle group,isovitamin A acid soft capsule group,combined treatment group and activator group.An acne model was constructed by injecting Propionibacterium acnes(C.acnes)into the auricle.Observe the apparent morphological changes of the mouse auricle and the swelling rate of the auricle.HE staining was used to observe the pathological changes of auricle tissue.The levels of serum inflammatory factors were detected by ELISA.The positive expression level of MMPs was detected by immunohistochemistry.Western blot was used to detect the expression levels of NLRP3/Caspase-1 signaling pathway-related proteins.Results Compared with the control group,the ear thickness,swelling rate,TNF-α,IL-6,IL-18 and IL-1β levels,MMP-1,MMP-3 and MMP-9 positive expression,NLRP3 and Caspase-1 protein expression levels in the model group were significantly increased(P＜0.05).Compared with the model group,the auricle thickness,swelling rate,TNF-α,IL-6,IL-18 and IL-1β levels,MMP-1,MMP-3 and MMP-9 positive expression,NLRP3 and Caspase-1 protein expression levels in the fire needle group and the isotretinoin acid soft capsule group were significantly decreased(P＜0.05).Compared with the fire needle group and isotretinoin acid soft capsule group,the auricle thickness,swelling rate,TNF-α,IL-6,IL-18 and IL-1β levels,MMP-1,MMP-3 and MMP-9 positive expression,NLRP3 and Caspase-1 protein expression levels in the combined treatment group were significantly decreased(P＜0.05).Compared with the combined treatment group,the auricle thickness,swelling rate,TNF-α,IL-6,IL-18 and IL-1β levels,MMP-1,MMP-3 and MMP-9 positive expression,NLRP3 and Caspase-1 protein expression levels in the activator group were significantly increased(P＜0.05).Conclusion The combined therapy of fire needle and isotretinoin soft capsule can significantly alleviate inflammatory lesions in acne model mice,and its mechanism may be related to the regulation of NLRP3/Caspase-1 signaling pathway to affect the levels of inflammatory factors and MMPs.关键词
