树鼩眼部多模态活体影像检测体系标准化构建及关键基线参数建立
Establishment of a standardized multimodal in vivo ocular imaging and assessment system in the tree shrew and derivation of key baseline parameters
摘要
Abstract
Objective To establish and standardize a multimodal in vivo ocular imaging and assessment workflow for the tree shrew and derive key baseline ocular parameters to support phenotyping and longitudinal follow-up in ophthalmic disease models. Methods Ten healthy adult tree shrews(five male, five female)were examined using a standardized protocol.Ocular volume was measured using B-scan ultrasonography, intraocular pressure(IOP)was assessed with a rebound tonometer, and the anterior segment was evaluated by slit-lamp microscopy. After pharmacologic mydriasis, color fundus photography, infrared fundus imaging, and fundus autofluorescence were performed.Fluorescein fundus angiography and indocyanine green angiography were conducted following intravenous administration of contrast agents to assess retinal and choroidal circulation.Optical coherence tomography centered on the optic disc was acquired for cross-sectional structural evaluation and retinal thickness measurements.Sex-related differences were analyzed. Results The eyes of the tree shrew are located bilaterally adjacent to the nasal bridge.Anterior segment structures were clearly visualized, with a distinct nictitating membrane. The cornea and crystalline lens were transparent, the iris was heavily pigmented, and the pupil was predominantly oval to nearly round.Fundus imaging revealed a centrally located optic disc and the absence of a macular fovea.Retinal vessels radiated outward from the optic disc in a spoke-like pattern, with alternating arterioles and venules, totaling approximately 10~16 primary vessels.Key baseline parameters were as follows:ocular globe volume,(0.262 0±0.004 5)mL;IOP:1 866.51~2 399.80 Pa(14~18 mmHg),mean:(2 153.15±103.99)Pa((16.15±0.78)mmHg)and retinal thickness,(250.36±2.90)μm.No statistically significant differences were observed between males and females. Conclusions We established a reproducible, standardized multimodal in vivo ocular imaging and assessment system for the tree shrew and generated baseline reference values for ocular globe volume,IOP and retinal thickness.Except for the absence of a macula, the ocular phenotype and several physiological parameters of the tree shrew are readily translatable to common clinical ophthalmic examination paradigms, supporting its utility as an experimental platform for disease modeling, therapeutic evaluation, and longitudinal in vivo monitoring.关键词
