Glucose and lipid metabolic dysfunction and cognitive decline:"metabolism-brain"correlation
摘要
Abstract
Dementia stands as a major cause of disability and mortality in the elderly population.Currently,no effective approaches are available to delay or treat dementia.Intervention at the early stage of cognitive decline(mild cognitive impairment)is considered as the most effective strategy to prevent the onset of dementia.However,there are several limitations,including an uncertain intervention time window,a lack of early blood/cerebrospinal fluid biomarkers,and no effective therapeutic medicines.Epidemiological evidence indicates that disturbances in central and peripheral glucose and lipid metabolism,primarily driven by diabetes,obesity and metabolic syndrome,serve as key drivers in the initiation and progression of cognitive impairment.Thus,systematically elucidating the epidemiological associations between dysregulation of glucose and lipid metabolism and cognitive decline,together with reviewing clinical and basic research findings and analyzing the complex molecular mechanisms network,and looking forward to key future research direction,holds significant importance for opening up new approaches to prevent metabolism-related cognitive impairment and reduce the incidence of dementia.关键词
