度拉糖肽联合运动疗法对2型糖尿病大鼠的影响
Effects of Dulaglutide Combined with Exercise Therapy on Type 2 Diabetic Rats

Abstract
Objective To investigate the improvement effect of dulaglutide combined with exercise therapy on type 2 diabetes mellitus(T2DM)model rats.Methods A T2DM rat model was replicated by feeding high cholesterol,high-fat diet,and intraperitoneal injection of streptozotocin.A total of 40 successfully modeled rats were randomly divided into the model group,dulaglutide group(dulaglutide 1.0 mg/kg),exercise therapy group(equal volume normal saline+autonomous running),and combination group(dulaglutide 1.0 mg/kg+autonomous running).A total of 10 normal rats were selected as the control group(equal volume normal saline).Rats in each group received subcutaneous injection of medication or normal saline once a day for 4 weeks.Blood glucose meter and sphygmomanometer were used to measure the blood glucose and blood pressure levels of rats,respectively.Morphology of the rats'heart and kidney tissues were observed by Masson staining method.Enzyme colorimetric method was used to detect the blood lipid levels;radioimmunoassay was used to detect insulin(INS)levels and insulin sensitivity index(ISI)was calculated;enzyme-linked immunosorbent assay was used to detect adiponectin,leptin,and fatigue related indicators.Results Compared with the model group,the fasting blood glucose,total cholesterol,and triglyceride in the dulaglutide group,exercise therapy group,and combination group were significantly reduced(P<0.05),as well as INS levels were significantly reduced and ISIs were significantly increased(P<0.05);the systolic blood pressure and diastolic blood pressure of rats in the dulaglutide group,exercise therapy group,and combination group were significantly reduced(P<0.05),while adiponectin levels were significantly increased and leptin levels were significantly reduced(P<0.05);the levels of nitric oxide synthase,inducible and native nitric oxide synthase were significantly reduced(P<0.05).Moreover,the indicators in the combination group were better than those in the single-treatment group.The diameter of myocardial cells in the model group was significantly larger than that in the control group(P<0.05),while those in the dulaglutide group and exercise therapy group were significantly smaller than that in the model group(P<0.05).The proliferation rate of glomerular mesangial matrix in the model group was significantly higher than that in the control group(P<0.05),while that in the combination group was significantly lower than that in the dulaglutide group and exercise therapy group(P<0.05).Conclusion Dulaglutide combined with exercise therapy can significantly reduce insulin resistance,improve hypertension symptoms,and alleviate fatigue in T2DM rats.关键词
