Abstract
Objective:To investigate the efficacy of dual-targeted therapy combined with paclitaxel and carboplatin as neoadjuvant treatment for human epidermal growth factor receptor 2(HER2)-positive breast cancer.Methods:A total of 88 patients with HER2-positive breast cancer admitted to our hospital from January 2022 to July 2025 were included.Stratified block randomization was used to assign patients to a control group(1 voluntary withdrawal;n=43)or a study group(2 voluntary withdrawals;n=42).The control group received standard treatment with dual-targeted therapy plus paclitaxel,whereas the study group received the same regimen with carboplatin added.Both groups were treated for 6 cycles.Efficacy was evaluated on the basis of radiologic response,the pathological complete response(pCR)rate,tumor marker levels[cancer antigen 15-3(CA15-3),carcinoembryonic antigen(CEA),and tissue polypeptide-specific antigen(TPS)],and safety.Results:After treatment,the objective response rate(ORR)was higher in the study group than in the control group(95.24％vs 79.07％,P<0.05),as was the pCR rate(90.48％vs 72.09％,x2=4.699,P=0.030).CA15-3,CEA,and TPS levels decreased in both groups and were lower in the study group than in the control group(P<0.05).No statistically significant between-group differences were observed in the grade distributions of anemia,neutropenia,thrombocytopenia,alopecia,rash,or nausea and vomiting(P>0.05).Conclusion:Under the conditions of this study,dual-targeted therapy combined with paclitaxel and carboplatin as neoadjuvant treatment for HER2-positive breast cancer increased the ORR and pCR rate and reduced tumor marker levels.关键词
