济生肾气丸"异病同治"尿石症和骨质疏松症机制探讨
Discussion on the Mechanism of Jisheng Shenqi Pills in"Treatment Different Diseases with Same Method"of Urolithiasis and Osteoporosis

Abstract
Objective To investigate the mechanism of Jisheng Shenqi Pills in treating urolithiasis and osteoporosis with the concept of"treatment different diseases with same method"using network pharmacology and molecular docking.Methods Active components and targets of Jisheng Shenqi Pills were screened from the TCMSP database,while disease targets for urolithiasis and osteoporosis were collected from OMIM;OMIM,GeneCards and DrugBank databases were used to obtain targets related to urolithiasis and osteoporosis,and then common targets of drugs and diseases were extracted.A protein-protein interaction(PPI)network of the intersection targets was constructed using the STRING database,and core targets were identified by the built-in algorithm of the cytoHubba plugin;GO and KEGG enrichment analyses were performed using the ClusterProfiler package in R 4.3.1 language to identify major biological processes and signaling pathways.A active ingredients targets-pathway network was constructed with Cytoscape software,and key active ingredients were screened based on network topology parameters;molecular docking was applied for validation.Results A total of 116 active ingredients of Jisheng Shenqi Pill were obtained,and 65 potential targets related to urolithiasis and osteoporosis were identified.Core targets included IL1B,MMP9,INS,IL6,TNF,PTGS2,etc.GO enrichment revealed that the main biological processes involved reactive oxygen species metabolic process,regulation of reactive oxygen species metabolic process and response to oxidative stress.KEGG enrichment indicated that major signaling pathways included the TNF,IL-17 and PI3K-Akt.Molecular docking demonstrated that the key active ingredients quercetin and kaempferol exhibited strong binding affinity with core targets.Conclusion The main active components of Jisheng Shenqi Pills,such as quercetin,kaempferol,diosgenin and wogonin,may active treating different diseases with same method of urolithiasis and osteoporosis through targets including IL1B,MMP 9,IL6 and TNF,by modulating signaling pathways such as TNF,IL-17 and PI3K-Akt,thereby regulating inflammatory responses and oxidative stress to realize the"same treatment for different diseases"of urolithiasis and osteoporosis.关键词
