金钱草及其易混淆品的特异性PCR鉴别
Identification of Lysimachiae Herba and Its Adulterants by Specific PCR

Abstract
Objective:To establish a specific polymerase chain reaction(PCR)method for the rapid identification of Lysimachiae Herba and its adulterants.Methods:Based on the differential loci in the ITS gene sequences between Lysimachiae Herba(original plant Lysimachia christinae)and its adulterants,specific identification primers for Lysimachia christinae were designed.DNA was extracted from various samples,and a PCR reaction system was established.The main factors affecting the PCR reaction were investigated,and the method was validated for applicability.The amplified products were detected by electrophoresis.Results:When the annealing tempera-ture was set at 60℃and the cycle number was 30,all samples of Lysimachia christinae showed a single,clear target band at 174 bp,while no such band appeared in the adulterant samples under the same conditions.Conclusion:The specific PCR method established in this study can rapidly and accurately identify Lysimachiae Herba and its adulterants,providing technical support for the authenticity and quality evaluation of Lysimachiae Herba medicinal material.关键词
