基于文献数据挖掘和网络药理学探究金钱草处方治疗高尿酸血症的用药规律及作用机制
Medication regularity and mechanisms of Jin Qian Cao-containing decoctions in the treatment of hyperuricemia:A study based on data mining and network pharmacology

Abstract
Objective:To analyze the formulation rules of Jin Qian Cao(Lysimachiae Herba)-containing Chinese herbal formulae for the treatment of hyperuricemia(HUA)and the mechanism of action of core drugs.Methods:Clinical research literature on Jin Qian Cao-containing Chinese herbal formulae for the treatment of HUA was retrieved from CNKI,Wanfang Data,and VIP database from their respective inceptions to October 31,2024.A database of Jin Qian Cao-containing prescriptions was established,and data mining was conducted through frequency analysis,binary association analysis,and cluster analysis.Drug pair combinations with a support degree of≥0.25 and a confidence degree of≥0.95 in the association analysis were considered as the core drug group of Jin Qian Cao.The intersection targets of the core drug group and HUA were collected through network pharmacology and submitted to the STRING platform for protein-protein interaction(PPI)network analysis.Cytoscape 3.7.0 network analysis was used to screen and visualize the core target PPI network model.GO function enrichment analysis and KEGG pathway enrichment analysis were performed on the intersection targets based on the Metascape database to predict the potential therapeutic mechanism of the core drug group on HUA.Results:A total of 112 effective prescriptions were included,involving 187 Chinese herbs.The most frequently combined herb was Fuling(Poria cocos);the main properties of the herbs were neutral and cold,the main flavors were sweet and bitter,and the main meridian tropisms were the liver and spleen meridians.The top three therapeutic effects of the herbs were water-draning and dampness-resolving,heat-clearing,and pain-relieving.The most commonly used couplet medicinal was Fuling-Jin Qian Cao.The association rule analysis suggested that"Jin Qian Cao-Fuling-Tu Fuling(Smilacis Glabrae Rhizoma)-Yiyiren(Coicis Semen)-Niuxi(Achyranthis Bidentatae Radix)-Bixie(Dioscoreae Hypoglaucae Rhizoma)-Huangbo(Phellodendri Chinensis Cortex)"was the core drug group of Jin Qian Cao-containing formulas.Cluster analysis resulted in three new formulae.Network pharmacology revealed that the potential core targets of the Jin Qian Cao core drug group for treating HUA were TP53,JUN,TNF,IL-6,etc.,and the main signaling pathways included AGE-RAGE pathway in diabetic complications and IL-17 signaling pathway.Conclusion:The main pathogenesis of hyperuricemia is the accumulation of dampness and heat.The treatment should focus on water-draning and dampness-resolving throughout the disease stages,and heat-clearing and pain-relieving should be added during acute gout attacks.The core drug group"Jin Qian Cao-Fuling-Tu Fuling-Yiyiren-Niuxi-Bixie-Huangbo"has a potential therapeutic mechanism for HUA by participating in inflammatory responses and regulating immune responses,providing a reference for the treatment of HUA with traditional Chinese medicine,prescription medication,and new drug development.关键词
