Abstract
Objective:Kaiyu Zhongyu decoction(开郁种玉汤)is a renowned classical prescription documented in the special chapter on"jealousy-induced female infertility"of FU Qingzhu Nvke(《傅青主女科》).This study aims to explore the network and potential mechanism of Kaiyu Zhongyu decoction in the treatment of"jealousy-induced female infertility"by network pharmacology.Methods:The Pharmacopoeia of the People's Republic of China(《中华人民共和国药典》)was used as thestandard for drug name standardization,and the natures,flavors and meridian tropisms of the Chinese herbal medicines in the formula were analyzed.The main active ingredients and targets of Kaiyu Zhongyu decoction were screened,and the relevant targets of jealousy-induced female infertility were retrieved.The drug-ingredient-target interaction network and protein-protein interaction network of Kaiyu Zhongyu decoction against jealousy-induced female infertility were constructed.Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed on target genes,and molecular docking was conducted subsequently.Results:A total of 34 main active ingredients and 947 predicted targets of Kaiyu Zhongyu decoction were screened out,along with 1 481 targets related to jealousy-induced female infertility,among which 117 common targets were obtained,including serine/threonine kinase 1(AKT1),interleukin-6(IL-6),tumor necrosis factor(TNF),tumor protein p53(TP53)and IL-1B.GO and KEGG enrichment analyses revealed that these targets were involved in multiple signaling pathways,including the advanced glycation end product(AGE)-receptor for advanced glycation end product(RAGE)pathway,phosphoinositide 3-kinase(PI3K)-protein kinase B(Akt)pathway,mitogen-activated protein kinase(MAPK)pathway and IL-17 signaling pathway.Moreover,core targets such as AKT1,IL-6 and TNF exhibited favorable docking activity with corresponding active ingredients.Conclusion:Kaiyu Zhongyu decoction exerts preventive and therapeutic effects on jealousy-induced female infertility via a multi-component,multi-target and multi-pathway integrated mechanism.Its various active ingredients regulate endocrine functions and cellular oxidative stress responses of the hypothalamic-pituitary-ovarian axis in females,thereby intervening against psychological jealousy-induced female infertility.关键词
