离子对反相高效液相色谱法测定暗纹东方鲀肉中呈味核苷酸
Ion-pair reversed-phase high performance liquid chromatography method for determination of flavor nucleotides in Fugu obscures muscle
摘要
Abstract
An ion - pair reversed - phase high performance liquid chromatography ( HPLC ) method for the determination of flavor nucleotides such as guanosine monophosphate (GMP), inosine monophosphate (IMP),cytidine monophosphate(CMP) and adenosine monophosphate(AMP) in Fugu obscures muscle was established.Chromatographic separation was performed on a SUPELCOIL LC-18-T C18 column at the wavelength of 254nm with the mobile phase of CH3 OH :0.01 mol/L KH2 PO4 solution (5:95, V/V) at 0.8 mL/min. The average recovery rate for four flavor nucleotides ranged from 96.70%~104.47% with the coefficient variance(CV) ranged from 1.12%~1.99% ,their minimum detection limits were 1.42,4.10,0.67,0.91μg/mL, respectively. The method was accurate and reliable,the operation was simple and easy.关键词
