山黧豆中还原型谷胱甘肽的荧光检测
Determination of reduced glutathione in Lathyrus sativus L.by fluorometric method
摘要
Abstract
A method for measurement of reduced glutathione(GSH) in grass pea( Lathyrus sativus L)tissues was developed,with use of o-phthalaldehyde(OPA) as a fluorescent reagent. By estimating the effects of pH, OPA contents and reaction time on the formation of fluorescent products at room temperature, the optimal assay conditions were achieved,including the excitation and emission wavelengths of 347nm and 425nm, respectively,pH8.0,0.30mg · mL-1OPA and a 20-min reaction time. The fluorescence intensity was linear with the concentration of GSH in the range of 0.05~0.80μg · mL-1 ,with regression equation y = 1084x +63.148 and correlation coefficient 0.9990.The method present was rapid,simple, sensitive and accurate,and more suitable for the analysis of large amount of grass pea samples.关键词
