Abstract

The immobilization of glucoamylase was carried out using glutaraldehyde as crosslink agent, using SBA-15 as carrier. The effect of immobilization temperature and time enzyme addition, pH value and glutaraldehyde concentration on immobilization result were investigated. The optimal immobilization conditions were determined. The results were as follows: the ratio of enzyme to carrier was 50 mg to 1.0 g, immobilization temperature 25 ℃, immobilization time 4h,pH 5.1, and the concentration of glutaraldehyde7.5%. Under these optimal conditions, activity recovery of immobilized enzyme was 56%. And the enzymatic properties of the immobilized enzyme were studied.The results showed that the optimal temperature of immobilized enzyme was 70℃, pH4.1, Km 0.022mol/L.The activity of immobilized enzyme was increased 18% after treated by ultrasonic. The best ultrasonic parameters obtained were as follows: ultrasonic power 100 W, temperature 70 ℃, and time 5 min.
