Abstract
A total of 89 Fusarium isolates were obtained from 45 diseased plants of maize top rot collected from some regions of Liaoning Province. Using traditional morphological classification and rDNA-ITS and EF-1a gene sequence analysis methods, 4 Fusarium species, Fusarium. subglutinans, F. verticillioides, F. proliferatum and F. oxysporum, were identified. Twenty-three isolates were clustered into 4 groups based on EF-1a gene sequence on phylogenetic tree. Additionally, genetic diversity of Fusarium species was analyzed using universally primed PCR (UP-PCR). The result indicated that a total of 57 bands appeared after amplification by using 5 primers, 53 bands (93.0%) of which were polymorphic. Twenty-four isolates were clustered into 4 groups at the similarity of 0.626 by UP-PCR cluster analysis. Our results indicated that EF-1a gene sequence could reflect the genetic diversity and differences of Fusarium species.关键词
