Abstract

The VB1 was extracted from fresh Chinese yam by ultrasonic extraction method, and the optimum HPLC conditions of VB1 in the Chinese yam were explored. A large number of experimental studies showed that the methanol: water = 5 : 5 as mobile phase,mobile phase flow rate at 1.0 mL/min, UV detection at 265 nm,column temperature at 25 ℃ was the optimum HPLC conditions of VB1 in the Chinese yam, the established standard curve had good linear relationship, the correlation coefficient R2 = 0. 998 9. Under these conditions, the average recovery of VB1 was 97.3%, relative standard deviation of the method precision less than 4.8%; the content of VB1 in the Chinese yam was 3.8 mg / 100 g. The method is a simple, fast, accurate and suitable for the analysis of VB1 in the Chinese yam.
