Abstract

The presence of Staphylococcus aureus in food represents a potential risk to public health, being its exterotoxins the major virulence factor. The purpose of this study was to develop a loop-mediated isothermal amplification (LAMP) method for the rapid, sensitive and simple detection of Staphylococcus aureus in food. LAMP primers were designed by targeting the nuc gene. With Bst DNA polymerase, the target DNA can be clearly amplified for 45 min at 65 degree C. LAMP products could be judged with agar gel or naked eye after the addition of SYBR Green I. The assay correctly identified Staphylococcus aureus, but did not detect 8 non- Staphylococcus aureus strains. In this way, 240 egg samples were investigated for Staphylococcus aureus. The accuracy of LAMP was shown to be 100% when compared to the standard method. Consequently, the LAMP assay is a sensitive, rapid and simple tool for the detection of Staphylococcus aureus and will facilitate the surveillance for control of contamination of Staphylococcus aureus in food.
