Abstract

ABSTRACT The separation and purification of difructose anhydride Ⅲ prepared by enzymatic synthesis using cation exchange resin DTF-02 was studied. The influence of column temperature,sample concentration,sample volume and elution flow rate were discussed. The study showed that,when 5mL difructose anhydride Ⅲ containing a soluble solid content of 20%,difructose anhydride Ⅲ was completely separated from fructo-oligosaccharide with elution flow rate of 1 mL/min at 60 ℃. Under such condition,the fractions containing DFA Ⅲ were separated and determined by HPLC which showed DFA Ⅲ content was over 95%. The research laid theoretical foundations for large-scale separation and purification of difructose anhydride Ⅲ.
