Abstract

Strain F is obtained, yielding 16 g/L γ-aminobutyric acid (GABA), from China traditional fermented vegetables. According to its colony morphology, physiological and biochemical properties and homology of 16S rDNA,strain F is identified as Lactobacillus brevis, and named as Lactobacillus brevis TCCC13007. The 30% positive mutation rate occurs when it is stimulated by diethyl sulfate and lithium chloride simultaneously for 17mins, and the mortality is more than 95%. After that, strains which grow in GYP agar medium with 50 g/L GABA inside are selected randomly, so that a 24 g/L GABA-yielding strain F1 is obtained by determining the content of GABA in fermentation culture. Its productivity is 50% more than the original and confirmed to be stable after an 8 times successive fermentation test.
