Abstract

Cross-linked enzyme aggregates (CLEAs) technique is a novel carrier-free immobilization method. In the present work, β-glucosidase from prune (Prunus domestica) seeds was immobilized by this method. The preparation conditions of CLEAs were optimized, and the synthesis of propyl β-D-glucopyranoside catalyzed by this immobilized enzyme was investigated. The optimum precipitant and its concentration, cross-linking agent (glutaraldehyde) concentration, cross-linking time were n-propanol, 2/1 (V/V), 10 mmol/L and 2 h, respectively. Under the optimum conditions, the activity recovery of n-propanol-CLEAs reached 84%. In the synthesis of propyl β-D-glucopyranoside catalyzed by the immobilized enzyme, the yield of the desired product was 60% after 96 h.
