啤酒中嘌呤的纯化及反相离子对色谱法测定的研究
Study on purification of purines in beer and determination of purines compounds by reversed-phase ion pair chromatography

Abstract
Four purines(Gua,Ade,Xan and Hyp)were separated and determined by reversed-phase ion pair chromatography.The optimized chromatography conditions were as following:254nm by UV detector with 0.02mol/L of KH2PO4 buffer solution added 1mmol/L 1-Hexanesulfonic acid sodium(pH=3.6 regulated by H3PO4 and methanol concentration=3%(v/v)regulated by quaternary pump)as mobile phase,and flow rate at 1.0mL/min.The average recoveries of four purines were 95.97%~101.74%,with the accuracy of the method as RSD of all four purines were 1.21%~2.04%.This method provided more feasibility than normal reversed-phase chromatography as to the purines including ion and non-ionic compound.The chromatography of beer sample was without interfering peak almost after beer sample purified by precipitation method,because of that determine the chemical composition and amount became simple and accurate.关键词
