褐藻寡糖分子量测定方法的研究
Study on the determination method of molecular weight of alginate oligosaccharide

Abstract
A rapid and accurate determination method of molecular weight of alginate oligosaccharide was studied. Alginate oligosaccharide A and B were degradated by oxidation and high pressure,and their molecular weight were determined respectively by ubbelohde viscometer and high performance liquid chromatography(HPLC). It was showed that,molecular weights of A and B were just 7300 and 5050,by ubbelohde viscometer,which were 10666 and 7935 however by HPLC. The molecular weights determined by two different methods coincided with each other but varied greatly. It was because that,the molecular weight determined by ubbelohde viscometer was calculated in empirical equation,not the absolute value;however,by HPLC,the linear regression equation was established between the log number of molecular weight(lgMw)and the retention time(t):lgMw=-2. 0338t+25. 834,R2=0. 9926,which returned to a good relation. In summary,HPLC was precise,efficiency,smart and simple,which was more suitable for the determination,and provided a theoretical and empirical basis for the molecular weight determination of alginate oligosaccharide.关键词
