淮山药薯蓣皂苷测定方法的研究

Abstract
Commonly used ultraviolet-visible spectrophotometry methods,vanillin-perchloric acid method,vanillin-sulfuric acid and methanol-sulfuric acid method,for determination content of dioscin were comparatively studied. The results showed that compounds formed under different treatments were very different at the maximum absorption wavelength and their stabilities. The methanol-sulfuric acid method was good,and the maximum absorption wavelength was 332nm,with good stabilities within 60min. The calibration curve was linear in the range of 2. 4~24μg/mL. Average content of dioscin in Dioscorea opposita Thunb was 0. 535mg/g with RSD 1. 06% by the methanol-sulfuric acid method. This method was simple,accurate,sensitive and stable,which could be used for the quantitative determination of total dioscin in Dioscorea opposita Thunb.关键词
