气相色谱法测定啤酒酵母β-葡聚糖含量
Quantitative analysis of β-glucan from brewers'yeast by gas chromatography

Abstract
In order to obtain an accurate content of β-glucan and mannosan prepared from brewers'yeast,a gas chromatography(GC) method was established through sequential steps of acetylation of hydrolyzed monosaccharide,option of chromatographic column,exploration of temperature program and drawing of standard curve.In addition,corresponding content in the product of Japan ADEKA joint-stock company was compared.The results showed that monosaccharide of β-glucan and mannosan could be separated well after acetylation.The purity of β-glucan and mannosan in product of this experiment reached 32.39% and 16.82%,respectively.However,the corresponding values in Japanese product came up to 30.63% and 16.13%,respectively.Finally,the results of repeatability and recovery experiments indicated that an accurate content of target polysaccharide could be obtained by using the GC method.关键词
