枝链霉菌L2001木聚糖酶的化学修饰及其活性中心
Chemical Modification and Active Site of Xylanase from Streptomyces Rameus L2001
摘要
Abstract
Many kinds of modifiers such as NBS.WRK and so on were used to react with xylanase from Strepto-myces L2001. The results showed that NBS can markedly lower the activity of the xylanase, while the reagents modifying glutamic acid, cysteine, lysine, arginine, serine and histidine residues had no significant effect on the activity of enzyme. The pseudo-first-order rate constant of the modified enzyme was measured for further studying the impact of tryptophan effect on the activity of enzyme. It was found that at least one tryptophan residue existed in the active site of xylanase from streptomyces L2001. The results also showed that tryptophan residue was at the binding sites of enzyme. Fluorescence spectroscopy of tryptophan indicated further that Tryptophan was in the enzyme's active site, which was essential to maintain activity groups.关键词
