微波辅助水解比色法快速检测羊奶粉中水解动物蛋白
Rapid Determination of Adulteration of Hydrolyzed Animal Protein in Goat Milk Powder by Microwave-Assisted Hydrolysis and Colorimetric Analysis

Abstract
Objective: To develop a fast and effective method for determining whether goat milk powder is adulterated with hydrolyzed animal protein(HAP).Methods: L-Hydroxyproline was used as an indicator of the existence of HAP.Samples were hydrolyzed under the assistance of microwave before colorimetric determination of L-hydroxyproline.The microwave-assisted hydrolysis process was compared with the conventional wet hydrolysis process.Results: The optimal microwave-assisted hydrolysis conditions of sulphuric acid concentration microwave power and hydrolysis time were 4 mol/L,720 W,and 45 min,respectively.Substantially identical results for the determination of L-hydroxyproline content in HAP and synthetic products adulterated with L-hydroxyproline were obtained from both hydrolysis processes with a RSD between-4.5% and 1.6%.The method allowed rapid determination of whether goat milk powder is adulterated with HAP,with high accuracy and reproducibility.Moreover,an excellent linear relationship was found between L-hydroxyproline concentration and absorbance with a determination coefficient of 0.9968.Spike recoveries between 86.24% and 91.30% were obtained.Thus,the requirements for the colorimetric determination of L-hydroxyproline were met.Conclusions: Microwave-assisted hydrolysis can shorten the sample pretreatment time from 16 h to 45 min.The method offers the advantages of rapidity,simplicity,good reproducibility and high recovery.关键词
