Effect of cold stress on membrane fluidity of Lactobacillus reuteri during freeze-drying

Abstract
The 1,6- diphenyl- 1,3,5- hexatriene (DPH) was used as fluorescence probe to label the membrane of Lactobacillus reuteri.The optimum conditions were established to measure the membrane fluidity of L.reuteri by labeling with DPH: excitation wavelength 360nm, emission wavelength 430nm, adjust ODs0o of L.reuteri to 0.8, incubateed at 30℃ in the dark for 20 min.Meanwhile,the survival rates,viability and membrane fluidity of L.reuten were measured after cold stress during freeze-drying.The results showed that the survival rates of freeze-drying Lactobacillus reuteri were improved by cold stress.At same time,the membrane fluidity was changed.The results implied that cold stress could improve cryotolerance of L.reuteri and prevent damage of freeze-drying.关键词
